<p>The chromatogram below illustrates&nbsp;the part-per-trillion analysis of a 1 ng/L mixed standard solution of PFAS (Per- and Polyfluoroalkyl Substances). Despite the extremely low concentration of 1 ng/L, the compounds are detected with good peak intensity. The LCMS-8065XE, equipped with the innovative StreamFocus and UFsweeper IV, delivers exceptional sensitivity for the analysis of trace-level components.</p>

<p>The LCMS-8065XE exhibits exceptional robustness during continuous analysis of complex matrices. The data above show both absolute peak areas relative to the initial response and peak area ratios normalized using internal standards, obtained from over 10,000 consecutive injections of treated wastewater samples. To create a more challenging analytical environment, matrix elimination using LC switching valves was deliberately omitted. Even under these challenging conditions, the system retained over 85% of its initial response after 10,000 injections, highlighting its exceptional durability and stability in demanding high-throughput applications.</p>

<p>The OAD-TOF system is a Q-TOF LCMS that realizes OAD (Oxygen Attachment Dissociation), Shimadzu&#8217;s proprietary fragmentation technology. It allows the analysis of the position of carbon-carbon double bonds in lipids and other organic compounds. Combined with the LCMS-9050, which boasts world-class high mass accuracy and fast polarity switching, it enables the creation of innovative applications.<br></p>



<h2 class="wp-block-heading">Innovative fragmentation technology</h2>



<p>The OAD-TOF system is equipped with an OAD RADICAL SOURCE I, which enables Oxygen Attachment Dissociation (OAD) analysis, an innovative fragmentation that utilizes Shimadzu’s proprietary radical reaction. It can measure fragment ions that cannot be obtained by conventional collision-induced dissociation (CID), where ions are fragmented by collision with an inert gas such as argon or nitrogen. The irradiating oxygen radicals specifically oxidize/dissociate double bonds between carbons, which is useful for structure estimation of organic compounds such as lipids. The technique can be applied to monovalent ions and negative ions, which have been difficult to fragment using radical reactions by electrons and anions, and provides completely new structural information</p>



<h3 class="wp-block-heading">What is OAD (oxygen attachment dissociation)</h3>



<p>CID preferentially dissociates weak chemical bonds. In the case of lipids, CID can determine the basic structure of lipids and polar groups, and the carbon composition of side chains (number of carbons and double bonds). With OAD, on the other hand, oxygen radicals react specifically with the double bonds between carbons, causing dissociation. By measuring fragment ions specific to the double bond, the position of the double bond can be determined.</p>

<ul class="wp-block-list"><li><strong>Radical source</strong><br>Neutral radicals such as O/OH/H radicals are generated by microwave discharge of raw material gases (water vapor and hydrogen gas) in a vacuum. The neutral radicals are introduced into the OAD cell through a quartz tube.</li><li><strong>OAD cell</strong><br>Precursor ions are reacted with neutral radicals generated in a radical source and fragmented. A collision gas (argon) can be introduced, and conventional CID fragmentation is also possible by setting the collision energy (CE).</li><li><strong>UF-FlightTube<img src="https://s.w.org/images/core/emoji/16.0.1/72x72/2122.png" alt="™" class="wp-smiley" style="height: 1em; max-height: 1em;" /></strong><br>Precise temperature control is achieved due to an optimized heater and heater sensor layout and a robust control system. This minimizes the effects of any room temperature variations to enable stable mass accuracy over long periods. Acquisition of a large amount of structural information by OAD and CID can lead to new discoveries in lipids and other organic compounds.</p>

<p>The OAD-TOF system can be used in combination with various options compatible with the LCMS-9050. This is especially useful for profiling lipids, where vast numbers of isomers exist.</p>

<p>Lipids in butter were analyzed using DPiMS in combination with the OAD-TOF system. Specific fragment ions derived from OAD were observed with only a simple pretreatment, which is a feature of DPiMS, and the double bond position of triacylglycerol was quickly estimated.</p>

<p>α-linolenic acid and γ-linolenic acid with different double bond positions were analyzed using Nexera UC and the OAD-TOF system. Isomers with similar structures were completely separated by SFC and detected with specific fragment ions derived from OAD, enabling highly accurate analysis. OAD can also be adapted to monovalent negative ions such as fatty acids.</p>

<p>The control software of the MUP-3100 automatically generates pretreatment results reports and stores them in the database along with pretreatment conditions. The MUP-3100 continuously monitors its operating performance, and if the device detects an error, its internal camera automatically creates a video of the 5 seconds before and after the occurrence, which is then saved along with error logs. This assists in identifying the root cause of errors and minimizing downtime.</p>

<h2 class="wp-block-heading">Processes 24 Culture Supernatant Samples Reliably in 6 Hours</h2>



<p>Conventional methods require complex operations to cut out glycans from antibody samples in the culture supernatant. The traditional method requires more than two days for preprocessing. When using the MUP-3100 and the antibody N-glycan analysis kit in combination, however, 24 samples can be preprocessed in six hours. Glycan profiling, including the type and quantity of glycans, can be performed by analysis of 2-AB labeled glycans using an HPLC fluorescence detector. The Shim-pack GIST-HP Amide [Metal Free] column is effective for separating out impurities and glycans contained in antibody preparations and culture supernatants.</p>

<p>The MUP-3100 can process 24 samples at a time. Even with simultaneous processing of other samples, the area ratio of each peak remains stable regardless of peak intensity, yielding data equivalent to that obtained when using the kit manually.</p>

<p>The LCMS-9050 is a quadrupole Time-of-Flight (Q-TOF) mass spectrometer system designed with technologies developed for Shimadzu LCMS series models and that results in among the highest mass accuracy stability levels available. Shimadzu’s proprietary high precision temperature control system inhibits even tiny mass variations caused by external factors, so that the system can be used to measure accurate mass values without worrying about mass calibration. The stable positive/negative high-speed polarity switching technology enables the simultaneous analysis of positive ions/negative ions, contributing to new applications development and heightened analysis efficiency. To ensure that all users can perform analysis easily and efficiently, Shimadzu has aimed to simplify mass calibration, maintenance and other non-analysis procedures. Further, combining this system with Shimadzu’s renowned lineup of varied optional equipment enables greater utilization of the characteristics of Q-TOF while greatly expanding the range of data obtainable.</p>



<h2 class="wp-block-heading">Simpler Accurate Mass Spectrometry</h2>



<h3 class="wp-block-heading">Technologies inherited from the Shimadzu LCMS series</h3>



<p>The LCMS-9050 provides excellent performance thanks to Shimadzu’s proprietary patented technology inherited from the LCMS series. The system is equipped with technology mainly inherited from the LCMS-8000 series, from the injection unit to the ion separator. Ions are efficiently injected into the TOF unit thanks to the bright ion optical system and high ion convergence. At the TOF unit, the precision temperature control system inhibits mass accuracy variations caused by external factors. The heater and temperature sensor are arranged at the optimal flight tube positions, providing highly accurate and robust temperature control. In addition, the electrical system controlling the flight tube is temperature controlled to achieve high mass stability.</p>

<h3 class="wp-block-heading">Stable long-term mass accuracy</h3>



<p>The precision-controlled temperature control system minimizes the effects of room temperature variations, so the LCMS-9050 provides extremely stable mass accuracy. For this reason, the system can be used to measure accurate mass values easily, without worrying about mass calibration. In addition, the time and effort expended to date on frequent mass calibration can be reduced.<br>Under usual laboratory conditions, six antibiotics (with molecular weights between 265 and 1203 Da) were consecutively analyzed, and the mass error against the theoretical values was plotted. When the compounds were analyzed three at a time in positive ion mode and negative ion mode, the mass accuracy was stable with accurate mass values for all compounds to within ± 2 ppm of the theoretical values without calibrating mass even once in 72 hours.</p>

<h3 class="wp-block-heading">Effortless tuning (Performance Assistant)</h3>

<p>A sub-probe separate from the main probe is included as standard with the LCMS-9050. The ionization unit is equipped with two probes, one for analysis and one for mass calibration, so the ionization unit does not need to be replaced during mass calibration or other equipment tuning. In addition, analysis and tuning can be performed without worrying about flow line contamination. Multiple sample vials can be stored in the unit, so the system can accommodate multiple calibration samples.</p>

<p>The LCMS-9050 includes an auto-tuning function that uses a standard sample to automatically adjust mass accuracy, resolution, sensitivity, and other parameters.<br>The function includes various tuning modes that can be selected depending on the instrument status, such as the full-tuning mode for tuning the entire system or other modes for tuning only the TOF unit for mass calibration. Auto-tuning can be executed from the LabSolutions<img src="https://s.w.org/images/core/emoji/16.0.1/72x72/2122.png" alt="™" class="wp-smiley" style="height: 1em; max-height: 1em;" /> LCMS workstation software without configuring any complicated settings, which means anyone can start an analysis with the system adjusted to its optimal state.</p>

<h3 class="wp-block-heading">Easy maintenance</h3>



<p>The system inherits easy maintenance from previous models, and is designed to simplify maintenance in the vicinity of the interface. The desolvation line (DL), which injects the sample into the vacuum unit, and the ESI capillary can be replaced quickly and easily. These parts can be replaced with the vacuum maintained as is, so system downtime can be minimized.</p>

<h2 class="wp-block-heading">Provides High-Speed Polarity Switching Even with TOF</h2>

<p>When switching between positive ion mode and negative ion mode, the flight tube voltage changes continuously to the target value. Mass errors will occur if analysis is performed before the voltage reaches the target value, so a stabilization standby time is required when switching polarity. With previous models, a certain amount of time was needed for stabilization standby, which made simultaneous positive ion/negative ion analysis difficult.<br>The LCMS-9050 provides high-speed polarity switching by substantially shortening the time required for stabilization standby during polarity switching. This is due to a high voltage power supply developed in-house by concentrating Shimadzu’s LCMS history and technology, and a proprietary polarity switching algorithm (UFstabilization, patent pending) developed by thoroughly investigating the properties of the high voltage power supply. Stable mass accuracy is shown even during high-speed positive/negative polarity switching, enabling simultaneous positive ion/negative ion analysis.</p>

<h3 class="wp-block-heading">Stability and high mass accuracy</h3>



<p>Just as with unipolar analysis, under usual laboratory conditions, six standard antibiotics (with molecular weights between 265 and 1203 Da) were subjected to simultaneous positive/negative ion analysis during a single analysis. The sub-interface was used to inject the external standard. Consecutive analyses were implemented until the external standard was used up, and a mass accuracy of within ±3 ppm was indicated for all compounds. Stable mass accuracy was maintained, even when performing long-term consecutive analyses using positive/negative ionization switching mode.</p>

<h3 class="wp-block-heading">A world of new applications</h3>



<p>With Q-TOF to date, when positive ion analysis and negative ion analysis were both performed, two measurements in total were required for the respective analyses, but with the LCMS-9050, the process is completed with a single measurement.<br>Analysis data accuracy is improved, because with pretreated samples under the same conditions, the positive/negative ions can be detected with no time difference. In addition, information on positive/negative ions can be obtained from a single analysis even for trace samples, which reduces the amount of reagents used. The data is output in a single data file, which makes it easy to compare the positive ion/negative ion analysis results.</p>

<p>A commercially marketed atorvastatin calcium sample was subjected to a simultaneous positive/negative ion analysis. With both positive ions and negative ions, the principle component atorvastatin and the impurities specified in the European Pharmacopeia (EP) were detected with high mass accuracy. Measuring in both positive ion and negative ion modes enables more accurate impurity identification.</p>

<h3 class="wp-block-heading">Eco-friendly</h3>



<p>The LCMS-9050 is recognized with the Eco-Products Plus label, Shimadzu’s proprietary eco-friendly product certification. Mobile phase and sample consumption can be reduced by 50 % by performing simultaneous positive ion/negative ion analysis, compared to when analyzing each mode respectively. In addition to reducing running costs during analysis, this contributes to the formation of a recycling-oriented society by promoting the efficient use of resources.</p>

<p>PESI features quick, convenient analysis without pretreatment. Exchanging the units takes a mere 15 seconds. This uses a probe to collect and directly ionize micro sample quantities, so there is no need for investigation of LC separation conditions or complicated sample pretreatment. Fluids and solids can be measured with only simple pretreatment. Data collection is complete in less than 1 minute even when detecting both positive ions and negative ions.<br>Combining this with the LCMS-9050 enables the identification of compounds in samples quickly and easily.</p>

<h3 class="wp-block-heading">Comprehensiveness of SFC × LCMS-9050</h3>



<p>A feature of supercritical fluid chromatography (SFC) is its comprehensiveness, which enables the separation and analysis of a wide range of compounds varying from low polarity to high polarity. Combining this with the LCMS-9050 enables the simultaneous analysis of positive ions and negative ions, so compounds with an even wider range of properties can be batch analyzed, heightening the comprehensiveness of a single analysis.<br>In addition, samples extracted by supercritical fluid extraction (SFE) cannot be reused, so in ordinary Q-TOF analysis, separately extracted samples are used for positive ion measurements and negative ion measurements. With the LCMS-9050 however, positive and negative ions are measured simultaneously, so information on both positive and negative ions is obtained from samples extracted under the same conditions.</p>

<p>The PESI unit can be removed easily. The ability to exchange it with an LC unit means measurements can be performed using the same theory of ionization and standard substance, which allows comparing data. The PESI unit was used for quantitative analysis of a forensic sample from a person that took the drug MT-45. As a result, we were able to obtain measurement data consistent with LCMS results in an extremely short time.</p>

<h3 class="wp-block-heading">Metabolomic Analysis Performed Easily<br>by Transferring LCMS Analytical Conditions to DPiMS Unit</h3>

<p>In this example, transition information for metabolites (26 components) such as amino acids, organic acids, and sugars were used for metabolomic analysis of mouse livers. Metabolomes can be analyzed easily by customizing some of the analytical conditions in the LCMS primary metabolite method package. The DPiMS-8060 system was used to measure the principal components in the model mouse group with acute liver damage induced by carbon tetrachloride and the control group. A significant difference was observed between model and control groups (given p &lt; 0.001 for Welch&#8217;s t-test), based on the significant contribution of taurine to the separation of groups in the PCA loading plot. The difference was verified with a box-whisker plot.</p>

<h3 class="wp-block-heading">Enables Use of LC/MS/MS Method Package</h3>



<p>The DPiMS-8060 system uses a direct analysis method that does not involve liquid chromatography using probe electrospray ionization. MRM parameters can be specified based on transition information included in the LC/MS/MS method package.<br>Consequently, methods can be created easily using an Excel template. A key feature of PESI is the easy process flow from pretreatment to measurement. For more detailed analysis, the ionization unit can be replaced with the ESI unit for measurements as an LC/MS/MS system.</p>

<h3 class="wp-block-heading">Data Analyzed Using LabSolutions LCMS</h3>



<p>The system can be used for a wide variety of applications, such as for library searches based on mass spectra obtained from product ion scans, for quantitative analysis based on ion chromatograms obtained from MRM, or for multivariate analysis using scan data.<br>Because the postrun analysis functionality in LabSolutions LCMS is used, data can also be easily compared to LC/MS/MS data.</p>

<p>Only a tiny amount of sample attaches to the probe during each sampling, reducing the risk of contamination inside the mass spectrometer to near zero.<br>Carryover can even be prevented by changing probes.<br>Even concentrated samples or samples of unknown concentration can be measured directly without pretreatment.</p>

<p>The qualitative analysis sequences for DPiMS QT and Q-TOF LC/MS were compared. Using DPiMS QT, it was possible to significantly reduce the measurement time in comparison to conventional methods using Q-TOF LC/MS.</p>

<p>Only about 15 seconds are required to remove the DPiMS QT unit, reconfiguring the system for Q-TOF LC/MS. The resources (solvents and columns) required for Q-TOF LC/MS analysis can be reduced by implementing qualitative analysis and primary screening via DPiMS QT, thereby reducing the number of samples requiring quantitative analysis.</p>

<p>PESI MS Solution software is used to control the probe, edit MS method files, and start measurements. Detailed MS analytical conditions are configured from the LabSolutions<img src="https://s.w.org/images/core/emoji/16.0.1/72x72/2122.png" alt="™" class="wp-smiley" style="height: 1em; max-height: 1em;" /> LCMS method editing window. Analysis can be started easily by selecting the probe control mode and MS method in PESI MS Solution. During analysis, the status can be confirmed in the LabSolutions LCMS window.</p>

<h3 class="wp-block-heading">Sequence for Qualitative Analysis via MS and MS/MS</h3>



<p>A qualitative analysis was performed on a whole blood sample mixed with trazodone (500 ng/mL). A cycle of MS and MS/MS measurements was performed simultaneously. LabSolutions Insight Explore was used for the data analysis. LabSolutions Insight Explore enables composition estimation, library searches, and structural analysis.<br>&nbsp;</p>

<p>The physical properties of target compounds in metabolomics can vary greatly. An analysis method tailored to the target compounds is therefore needed for comprehensive measurements of metabolites.<br>This database contains multiple ready-to-use methods for comprehensive analysis of metabolites with different properties. Pretreatment examples are also included for various metabolites and samples. The user can begin a metabolomics analysis without time-consuming evaluation of LC or MS conditions.</p>



<h2 class="wp-block-heading">Information for 470 compounds</h2>



<p>This database is based on the successful LC/MS/MS Method Packages for Primary Metabolites, Cell Culture Profiling, Lipid Mediators, Short Chain Fatty Acids, and Bile Acids, and provides a comprehensive list of 470 metabolites (including internal standards) with retention times and exact mass information.</p>



<p>The database is compatible with the Multi-omics Analysis Package (sold separately). Processing the analysis results with Multi-omics Analysis Package allows statistical analysis, network analysis and visualization on the metabolic map(exc. Short chain fatty acids) to be carried out easily.​</p>
